Abstract (EN)

Dilated cardiomyopathy (DCM) is a progressive disease of heart muscle characterized by contractile dysfunction and ventricular dilatation with normal left ventricular wall thickness. There are many causes of cardiomyopathy including hereditary diseases, infections and toxins. According to echocardiography and pedigree studies; approximately 20% of DCM cases are assigned a diagnosis of idiopathic dilated cardiomyopathy. Disease could be seen in the other members of family due to famial inheritance. The patient's other family members also (including children) are required to be checked their health conditions. During the treatment of DKMP; the resistance mechanisms and side effects of drugs, varies between individuals in society due to genetic variations in drug metabolizing enzymes. This difference may be depends on the expression of MRP genes that makes the multiple drug resistance. In this study, as a result of the evaluation of gene expression analysis of MRP-1 and MRP-2, we aimed to explain the differences of the treatment response of the DCM patients, who need the multiple drug usage, and help to clarify the molecular bases of treatment. 90 DCM patients and 90 healthy individuals peripheral blood samples were collected for total RNA isolation. cDNA was prepared by using the total RNA isolated from the samples. Q-PCR was evaluated by using the target genes (MRP-1 and MRP-2) and a reference gene (ABL) primers, the reference and target cDNA's were replicated. The expression levels of the target gene and the references were measured for analysis. According the results of the analyses, the DCM patients had 6 times increased expression levels on MRP-1 gene mRNA versus healty individuals and 77 times increased expression levels on MRP-1 gene mRNA versus healty individuals. Evaluation the gene expression of MRP-1 and MRP-2 may contribute to the treatment of DCM disease. But this study needs to be improved by icreasing the gene families and supported by the other scientific data.
