Abstract (EN)

THE EFFICACY OF PROBIOTIC MICROORGANISMS AGAINST ENTEROINVASIVE PATHOGENES IN CELL CULTURE Introduction and Objective: Probiotics were described as "living microorganisms which help to keep the host healthy when taken in sufficient amount." For this reason, research has been carried out for its use in a wide variety of fields including both therapeutical and prophylactical fields. Although it is known that gastroenteritis caused by enteroinvasive pathogens is treated with antibiotics, the negative effect of antibiotics produced on the flora and the resistance problem developing against antibiotics in microorganisms brought the search of new treatment into question. Materials and Methods: The present project aimed to investigate the inhibitory roles of probiotics against pathogenic bacteria in in vitro environment and to improve the treatment strategies against pathogenic enteric bacteria using probiotics. In the present study, Lactobacillus acidophilus, Saccharomyces boulardii and artificial colonic epithelial cells were used in order to elucidate the mechanisms of action of probiotics against Salmonella Typhi and Shigella dysenteria which cause intestinal epithelial damage. Results: In the first step of the study to understand the efficacies of probiotic microorganisms and soluble products on pathogenic microorganisms, it was observed that probiotics did not produce an inhibition zone against pathogenic microorganisms in the Müller Hinton plaques. In the wound healing cell proliferation division, which is the second step of the study, it was observed that S. Typhi and S. dysenteria impaired the wound healing cell proliferation process of the Caco-2 cells in the cell culture, that L. acidophilus alone had no effect on this proliferation, however, S. boulardii alone increased the proliferation of damaged cells compared to the control group. In the infection model, it was examined whether the pathogenicity of S. Typhi and S. dysenteria which are known for their destructive effects decreased or not. When examined in the accompany of L. acidophilus and S. boulardii, they were observed to accelerate the wound healing compared to the combination of L. acidophilus with S. Typhi whereas it did not have any effect on wound healing in case of its combination with S. dysenteria. Whereas S. 61 boulardii could not impede the destructive effect created by the equal amount of S. Typhi and S. dysenteria and currently used bacterial amounts of it. Conclusion: The effect of probiotics on acute infectious, invasive bacterial diarrhea is not clear. Despite the significant statistics concluded, there is a need for more randomized controlled trials in order to understand whether they have clinical significance or not.
