Identification of significant viruses in cucurbits produced in Hakkari province

Abstract (EN)

In this thesis study; Cucumber mosaic virus (Cucumber mosaic virus; CMV) and Zucchini yellow mosaic virus (ZYMV) which are viral agents that cause harmful and economic losses in cucurbits in Hakkari province investigated serologically by DAS-ELISA (Double Antibody Sandwich Enzyme-Linked Immunosorbent Assay) method. It has been reported by studies that these viral factors cause serious economic losses worldwide. Leaf samples were collected from cucurbits, which exhibit yellowing, necrosis, mosaic, and deformity symptoms on leaves suspected of virus disease from Kırıkdağ, Otluca, and Merzan areas in Hakkari. DAS-ELISA test was used to identify viral agents in a total of 92 leaf samples from 7 watermelon (Citrullus lanatus L.), 5 melon (Cucumis melo L.), 38 cucumber (Cucumis sativus L.), 15 zucchini (Cucurbita pepo), 20 pumpkin (Cucurbita moschata) and 7 acrid (Cucumis melo var. Flexuosus) showing viral symptoms was applied where cucurbits are grown in Hakkari, with a survey study carried out in September 2022. While CMV was detected in 29 of 92 samples by the DAS-ELISA test, ZYMV could not be detected in any sample. CMV, which was detected at a rate of 31,52%, was most commonly detected in Otluca with the number of infected samples. In Cucurbit cultivars, a rate of 85,71% in 6 of the 7 samples tested were detected in acrid. Key Words: Cucurbits, CMV, ZYM, DAS-ELISA, Hakkari
