Determination of relationship between HSP90 and ovarian and endometrial cancer

Abstract (EN)

Ovarian cancer constitute 25% of genital cancers in women and its referred as the second most common gynecological cancer in developed countries. According to the data of the Ministry of Health in 2002, the most lethal of gynecological cancers and it has been identified as within the fourth lathal in women's cancers in our country. Endometrial cancer determined the most common malignant tumor of the female genital tract and 75% of patients is postmenopausal who is generally applied in the presence of symptoms presenting with stage 1. Although there is no effective screening test, due to a symptomatic in 75% of cases can be diagnosed at an early stage. Heat shock protein is protein cluster which is strongly stimulated with various agents as heat shock, physical stress. Except this feature, hsp regulate structure of proteins and modifying as molecular chaperone. Heat shcok factor (HSF) which is transcription factor, regulate heat shock protein gene trancripition. HSF effect as initiator transcription activation. There is two major isoforms in cytoplasm in HSP90. HSP90α is induclable form and HSP90β is constituve form. While HSP90α is stimulated in stress conditions, HSP90β is aktive fom as constituve protein. Although mutant genes involved in cancer cells of proteins is stabilised via HSP90, chemotherapeutic drugs have been developed for level of HSP90 in cell and modifing of HSP90 capacity. In our studies, we aimed to investigate the role of HSP90AA1, HSP90AB1 and HSP90B1 polymorphism in ovarian cancer (n=77), endometrial cancer (n=13) and healty control (n=44). In our study we found that in terms of HSP90AA1 and HSP90B1 gene polymorphism is not statistically significant in cases and control. In terms of HSP90AB1 polymorphism, while between ovarian cancer and control group is not statictially significant, CC genotype is higher in endometrial cancer than control grup. Also T allele of HSP90AB1 is higher frequency in ovarian cancer higher than control group. The result of this study carries the attribution of being the first data on the metioned gene sites in ovarian cencer and endometrial cancer in Turkish population.
