Abstract (EN)
Objective: The aim of this study is to generate the cytotoxic Panton-Valentine lökosidine (PVL) using recombinant technology and to examine its effect on the prostate cancer cell line. Materials and Methods: Primers to amplify LukS-PV and LukF-PV genes separately and together were designed according to genebank (NCBI CP084892), Amplified fragments were cloned into the pET30a(+) expression vector. Cloned fragments were confirmed by sequence analysis and used to obtain recombinant proteins. The synthesized proteins were purified partially and their effects on macrophages were investigated. Then, the cytotoxic effect of LukS and LukF seperatelly and together were tested on the normal prostate cell line (RWP-1) and prostate adenocarcinoma cell line (PC3). Cytotoxic effects were measured with the WST-1 test and the results were evaluated. Results: Recombinant LukS-PV and LukF-PV and in combination were obtained. Then, their effects on macrophages were examined, it was seen that rPVL and LukS-PV had cytotoxic effect on macrophage, but LukF-PV had no effect. When applied to prostate normal cell line (RWP-1) and adenocarcinoma cell line (PC3), the recombinant products obtained did not have any cytotoxic effect on either cell line. Conclusion: Although Panton Valentine Lökosidin had sitotoxic effect on macrophage cell line, no sitotoxic effect was observed on prostate normal cell line (RWP-1) and adenocarcinoma cell line (PC3).
