Abstract (EN)

Are bacteria K. pneumonia from the existing natural flora inside Gutwhereit causes many diseases and of which (Urinary tract infections and pneumonia Pneumonia septicemia Septicemia) And increased interest bacteria Klebsiella pneumoniae is an opportunistic pathogen and has several factors that help it cause disease, such as having a capsule that resists phagocytosis and is responsible for the high incidence of acute diseases caused by it. These bacteria are urease-positive and have the ability to form Biofilm. Urinary tract infection is the most common type of bacterial infection and untreated it can lead to serious losses to humans. The study included the collection of 104 urine samples from women and men with urinary tract infection from the auditors of Salah al-Din General Hospital and the primary health care sector in the city of Tikrit for the period 12/15/2021. Until 15/4/2022, urine samples were collected from the consultative laboratory and transferred to the bacteriological laboratory in the city of Tikrit. Then the investigation was conducted to investigate the pathogens of urinary tract infections, diagnose them, and test their sensitivity to antibiotics. The results of the bacterial culture showed that 78 urine samples gave positive growth at a rate of 75% and showed bacterial growth. Klebsiella pneumonia is Gram-negative with 55% of samples. The isolated bacteria were diagnosed by microscopic, culture and biochemical methods, and the diagnosis was confirmed by examination with Vitek compact 2 system. The results showed that women are more infected than men. Get an antibiotic sensitivity test Using the disc spread method For (10) different antibiotics, some of them have multi-antibiotic resistance, with differences in the pattern of resistance between bacterial isolates.The results of the antibody susceptibility test showed that the antibody Cefotaxime, Trimethoprim and Gentamicin are the most effective Inhibitory direction of the isolated bacteria As all isolates were 100% sensitive to them, while The results showed that the rest of the antibiotics used in the study were moderately sensitive The direction of the isolated bacteria, which are antibacterial Tetracycline, Cephalexin, Azythromycin, and 70%. While the results showed that the antagonist Rifampin showed the lowest sensitivity to most bacterial isolates, with a percentage of 20% The results have been shownour study Ban the antidote Amoxicillin and Erythromycin and Vancomycin used in the study were all 100% resistant to it. Bacterial isolates have been identified pneumoniae K molecularly using a PCR amplification chain reactionusing qualitative prefixes (specific primer) to detect the presence of virulence factor genes as the genes amplified rRNA S 16, rmpA genes and K2wzy genes, as well as gene amplification using multiplex chain amplification reaction. The results showed that all isolates of Klebsiella pneumonia gave a clear bundle with a size of200Husbandbasicand volume500 base pairsand size650 base pairs When using specialized primers for genes. The results of sequencing the nitrogenous bases of technical products showed PCR for the samples under study matches up to (80%) with the sequence of nitrogenous bases of the virulence factors genes present in the studied bacterial isolates that cause bacterial resistance.
