Abstract (EN)

In this researh, DNA barcoding of Sideritis trojana Bornm. (Lamiaceeae) and Digitalis trojana Ivan. (Scrophulariaceae) which are endemic plants of Ida Mountain were investigated. DNA barcoding process were realized in the matK (Maturase) gene and trnH- psbA intergenic spacer. DNA extracted from the young leaves of endemic plants, PCR amplifications were performed according to the matK (Maturase) gene and trnH- psbA intergenic spacer regions? primers. Concerned PCR zones were observed with agarose gel under UV. The PCR products were sent to MACROGEN company for sequencing. After the sequences were obtained from MACROGEN company, some bioinformatics programmes were used to to compare the classical taxonomical classifications of species with the molecular classifications of the species gathered by DNA barcode based trees. Possibilities of being core barcode region of matK (Maturase) gene and trnH- psbA intergenic spacer were discussed and the relatives of Ida Mountain endemic plants were evalutated with the other plants taken from BLAST. Multiple Sequence Alignment of sequences were performed by using CLUSTALW2 and T-COFFEE programmes. And totally 43 plant sequences were taken from BLAST programme for aligning of these sequences with Ida Mountain endemic plants for DNA barcoding. Neighbor joining method and Kimura two- parameter was used from MEGA 4.1 for obtaining DNA barcode based trees.According to the DNA barcode based tree classifications, molecular classification of species observed consistent with the classical taxonomic classifications of species. matK was more suitable for the plants used in this investigation because of the easiness of the alignment and gathering the tree. It was generally enough to discriminate the species but combination of matK with more variable region can be suggested in the identification-based DNA barcoding investigations.Keywords: Sideritis trojana, Digitalis trojana, matK, trnH- psbA, DNA barcoding.
