Abstract (EN)

Histological Changes After Boron and Vitamin B17 Application in Methotrexate Hepatotoxicity and Evaluation of Serum Thiol-Disulfide Balance Objective: Methotrexate (MTX), a folic acid antagonist, is a chemotherapeutic drug used in the treatment of various inflammatory diseases as well as some types of cancer. The purpose of this study; To examine the effects of boric acid (BA) against the hepatotoxic side effect of MTX at light microscopic and biochemical level. Materials and Methods: There will be 6 male Wistar Albino rats in each group way; control was divided into 9 groups as MTX, BA, B17, BA + B17, MTX + B17, MTX + BA, BA + MTX, MTX + BA + B17. On the 11th day of the experiments, liver tissues of rats in all groups were removed rapidly by midsagittal incision after decapitation performed under anesthesia (75mg / kg ketamine + 10mg / kg xylazine) by intraperitoneal (i.p) application. It was fixed with 10% formaldehyde. Paraffin blocks were prepared and 4μm thick sections were cut from the blocks. It was stained with Hematoxylin & Eosin (H&E) and Masson Trichrom stain. It was examined and photographed with an Olympus BX43 photomicroscope. Scoring was done using the Score method. In addition, various oxidant (disulfide,% disulfide / native thiol, MPO, LOOH) and antioxidant (native thiol, total thiol, catalase, albumin) liver function tests were measured in the sera of the rats. All biochemical tests measured in the study were performed on the Siemens ADVIA 1800 Auto Analyzer (siemens healthycare GmbH, Erlangen, Germany). Results: Light microscopic examinations revealed sinusoidal dilatation, hepatocyte degeneration, vascular congestion-thrombosis, and inflammatory infiltration in the liver of rats given MTX. It was observed that the sparing of boric acid was more effective than its treatment. In the groups given MTX, the level of oxidative stress-related parameters such as lipid hydroperoxide, MPO and disulfide increased, whereas the level of antioxidant parameters such as native thiol, total thiol and catalase decreased. Conclusion: In this study, it was determined that the protective effect of boric acid was more than the therapeutic effect in liver damage caused by methotrexate. Oxidative hepatotoxicity caused by methotrexate disrupted the thiol disulfide balance and caused it to shift in favor of oxidation. In the light of our findings, it was predicted that administering amphipathic thiol donors (alpha-lipoic acid, etc.) with both lipophilic and hydrophilic properties to patients before starting MTX treatment would significantly reduce the side effects of MTX.
