Abstract (EN)
In this study, the effects of growth regulator combinations on micropropagation of OHxF 97 and OHxF 333 pear clone rootstocks were investigated. In the proliferation phase of the research, the effects of different doses of BAP, a growth regulator, sucrose and glucose as a carbon source, and silver thiosulfate were investigated. In the first trial of the proliferation stage, it was determined that the addition of 2 mg/l BAP + 0.5 mg/l IBA to MS nutrient medium gave the highest shoot number per explant (5.2 and 2.1 units/explant, respectively) in both OHxF 97 and OHxF 333 rootstocks. In the second experiment of proliferation, the effects of carbon source and silver thiosulfate on shoot number per explant and shoot length in OHxF 97 rootstock were found to be insignificant. It was determined that the addition of glucose and glucose + 5 µM silver thiosulfate to the nutrient medium in OHxF 333 rootstock significantly increased the number of shoots per explant (2.6 and 2.5, respectively) compared to the control. In the rooting stage, the effect of combinations of IBA and activated charcoal was determined in the first trial. In the study, the highest rooting rate occurred in ½ MS medium containing 2 mg/l IBA without activated charcoal on both rootstocks. It was determined that the addition of activated charcoal to the nutrient medium decreased the rooting rate compared to the control. In the second experiment, the effect of 2 mg/l IBA + 100 or 150 mg/l organic iron (Fe-EDDHA) application was investigated. In the second stage of the rooting study, the highest rooting rate was achieved in ½ MS medium with 2 mg/l IBA + 150 mg/l iron added in both rootstocks. It was determined that the addition of organic iron to the nutrient medium increased the rooting rate.
