Abstract (EN)

Oligostenoteratozoospermia is a condition characterized by the deterioration of sperm count (oligozoospermia), motility (asthenozoospermia) and morphological structure (teratozoospermia) that causes reproductive problems in men. Mitochondrial dysfunction and the resulting oxidative stress play an important role in the pathogenesis of this disease. Mitochondria are the energy centers of sperm cells and are vital for their motility and fertilization abilities. Oxidative stress can damage lipids, proteins and DNA in sperm, leading to reduced fertility. Recently, intensive research has been conducted on the protective effects of natural substances, especially antioxidant polyphenols, against this type of molecular damage. One of these, resveratrol, is a natural polyphenol found in grape skins, raspberries, peanuts and some plants, and is known for its strong antioxidant, anti-inflammatory, antiapoptotic and cell-protective effects. Studies have shown that resveratrol supports mitochondrial function, reduces the formation of reactive oxygen species (ROS) and plays an important role in maintaining cellular energy balance. The aim of this thesis is to investigate the effect of resveratrol on sperm mitochondrial stress and apoptosis in oligostenoteratozoospermic individuals. Within the scope of the study, sperm samples taken from men diagnosed with oligostenoteratozoospermia were washed with a solution containing resveratrol under laboratory conditions and then mitochondrial membrane potential and DNA damage, apoptosis and sperm motility and morphology parameters were evaluated using various histological analysis methods. This thesis examines the scientific basis of the use of resveratrol as an alternative and adjunctive agent in the treatment of male infertility, offering new directions for the development of natural intervention strategies in clinical practice. The results of this study revealed that the addition of resveratrol to washing solutions does not play a role in stabilizing mitochondrial activity in sperm and normalizing energy metabolism. It has also been shown that additional applications of Resveratrol to sperm washing solutions may change dose-dependently.
