Abstract (EN)

vu SUMMARY Methods were developed for organogenesis of pecan, "Carya illinoensis" using tissues of mature seeds and lateral, apical buds of mature trees. Effective surface sterilization methods were achieved for the production of sterile explants from mature seeds and mature lateral, apical buds oîCarya illinoensis. Methods were developed for organogenesis of kiwifruit (Actinidia deliciosa) using tissues of mature seeds and plant regeneration from leaf callus. Mature seeds of pecan were cultured in MS medium containing different concentrations of BAP and kinetin (0.5 - 4.0 mg l"1). Best results for germination was obtained with 2.0 mg l^BAP concentration. Apical and lateral buds from mature trees of pecan have been cultured on MS medium containing different concentrations of BAP and kinetin (0.5 - 4.0 mg l"1), for a period of one year. Buds (lateral and apical) were not developed on these concentrations during tins period. Effects of explants source, strength of MS, types of sugar and BAP concentrations on axenic germination of seeds from kiwi were examined and 2.0 mg l"1 BAP, 30 g/1 sucrose and 1/4 MS were found to be the optimum medium. Effect of the strength of MS, types of sugar and cytokinin on shoot proliferation of kiwi were investigated. The optimum result for shoot proliferation was obtained on 1/1 MS medium containing 0.5 mg l"1 BAP and 30 g/1 sucrose. The shoots developed m this medium were rooted on MS medium with 1.0 mg l"1 NAA. The in vitro rooted shoots were succesfully adapted at in vivo condition. Effects of explants source, auxin (1.0 mg l"1 NAA) and cytokinin (1.0 mg l"1 BAP) supplemented with 2.0 mg l'^^-D on the initiation of callus were examined m order to obtain callus for plant regeneration. The best callus for plant regeneration was obtained from leaf explants on MS medium containing 1.0 mg l"1 NAA + 2.0 mg T^^D. After several subcultures, the shoots were obtained from callus m all MS media containing different BAP concentrations (2.0 - 8.0 mg l"1) tested. The best shoots formation were obtained on MS medium containing 4.0 mg l"1 BAP.
