ABSTRACT IN EXPERIMENTAL KIDNEY ISCHEMIA REPERFUSION INJURY ON RATS ELABELA, APELIN-13, CYSTATIN-C AND NITRIC OXIDE LEVELS AND THE PROTECTİVE EFFECT OF N-ACETYLCYSTEINE The aim of this study was to look at and compare the levels of ELABELA, Apelin 13, Cystatin-C and Nitricoxide in renal ischemia / reperfusion injury in rats and to determine the effects of N-Acetylcysteine on ischemia / reperfusion by histologically and by the mentioned molecules. The present study was performed on 8 weeks 68 male Wistar-albino male rats obtained from Firat University Experimental Research Center in Firat University Experimental Research Center (FÜDAM) and Firat University Faculty of Medicine Histology Embryology Department Laboratory. The subjects were randomly divided into 7 groups. Groups are determined as Group 1 (control group) (n = 8), Group 2 (Sham Group) (n = 10), Group 3 (Unilateral Renal I / R group) (n = 10), Group 4 (Bilateral renal I / R group) (n = 10), Group 5 (NAC group with Unilateral Renal I / R) (n = 10), Group 6 (NAC group with Bilateral Renal I / R) (n = 10), Group 7 (NAC Group) (n = 10). When the groups were examined, no significant difference was observed between Control, Sham and Group 7 in terms of biochemical and histopathological features. Urea and creatinine levels were found to be increased in Group 4 and 6 compared to the control group. In group 3, while there was no significant increase in urea but a significant increase in creatinine level was detected according to the control group. In group 4, urea and creatinine levels were significantly higher than Group 3. No significant difference was found between Group3 and Group 5 according to urea and creatinine levels. In Group 6, no significant difference was found in urea and a statistically significant decrease in creatinine value was determined according to Group 4. According to the control group, significant decrease in TAS levels and increase in TOS levels were found in Group 3, Group 4, Group 5 and Group 6. In Group 4, TAS, ELA and Apelin-13 decreased significantly compared to Group 3; while significant increase in TOS, Cystatin-C and INOS levels were detected. In Group 5, there was a statistically significant increase in TAS, ELA and Apelin-13 levels according to Group 3. In Group 6 according to Group 4 significant reduction in creatinine, TOS, cystatin-C and INOS; significant increase in TAS, ELA and Apelin-13 was found. As a result, in this study, it was seen that ELA and Apelin-13 decreased and significantly increased INOS and Cystatin-C secondary to ischemia. Urea and creatinine were evaluated as not as specific as this molecüles in ischemia reperfusion injury. After N-acetylcysteine treatment, both INOS and Cystatin-C decreased with the increase in ELA and Apelin-13 following improvement in both tissue and serum samples. Studies of these molecules, especially Apelin-13 and ELA, are used in the treatment, and it is thought that the increase in these studies will shed light on the early detection and treatment of renal ischemia and reperfusion injury.
