Abstract (EN)
The encephalopathy which develops following sepsis is defined as life-threatening organ failure due to the irregular response of the host to infection which has extremely high mortality-morbidity rate and affects the nervous system and muscle. In our study, we aimed to investigate effects of inflammation on brain and muscle tissue and the effects of GABAA receptor antagonist Bicuculline in rats with sepsis. Experimentally, sepsis induced with LPS on rats; they were seperated 4 groups: control, LPS (10 mg/kg i.p.), Bicuculline (1.5 mg/kg s.c.), LPS+Bicuculline. Electrophysiological recordings and body temperature measurements were completed at 24 hours after injection, Blood was taken from the heart for Leukocyte count and Neuron specific enolase (NSE) levels measured. Brain and gastrocnemius muscle tissue taken for TNF-α and IL-10 and GABA levels were measured by ELISA method. Tissue imaging was performed with S100-ß, NEUN and Sinaptophysin by immunofluorescence staining. One-way ANOVA and Tukey test was used for statistical analysis. In our results, we observed in LPS group inflammatory parameters were significantly increased in brain and muscle compared to the control and caused S-100ß imvolvement increased and synaptophysin involvement decreased in immunofluorescence staining resulting in tissue level damage. In electrophysiological recordings, activity consistent with acute non-focal seizures was observed in the LPS group, while it was consistent with resting status in others. We thinks that Bicuculline Methiodide that is a GABAA antagonist, may be a prophylactic agent in the sepsis which caused to impaired neurotransmitter balance, increased pro-inflammatory cytokine and lipid peroxidation, decreased anti-inflammatory cytokine levels.
