Myd 88 and tlr-9 polymorphism in children diagnosed with sepsis

Abstract (EN)

Aim: We aimed to investigate whether sepsis had genetic tendency or not. Therefore we studied MyD 88 and TLR-9 polymorphisms which had important roles on the immune system in children diagnosed with sepsis.Material and methods: 130 patients, 65 of whom were diagnosed with sepsis in study group and 65 of whom were without sepsis in control group, were included between April 2010 and January 2012 in Pediatric Intensive Care Unit in Çukurova University. The demographic characteristics, including PIM II, PRISM II scores, underlying diseases, cultures, organ failure, the treatment modalities and the treatment results were recorded. After DNA had been obtained from blood samples in study and control group, polymorphisms of MyD 88 and TLR-9 were analyzed. According to the genotype and allel frequencies, the distributions of polymorphisms of MyD 88 SNP -938 C/A (rs4988453), MyD 88 SNP 1944 C/G (rs4988457), TLR-9 SNP 2848 A/G (rs352140) and TLR-9 SNP 1237 C/T (rs5743836) were analyzed in both study and control group.Results: Median age (p=0.254) and gender (p=0.724) were not statistically significant in study group. Statistically C/C genotype of MyD 88 SNP -938 were more signficant than that of C/A genotype in patient group (p=0.002). No statistically significant difference was found between study and control group in terms of MyD 88 SNP 1944 genotypes, TLR-9 SNP 2848 genotypes and TLR-9 SNP -1237 genotypes (p=0.272, p=0.921, p=0.232, respectively). The mortality was 23,07 % in patients with sepsisConclusion: We assume that the studies related with gene polymorphism could have a beneficial role in order to the determining of sepsis tendency and management of treatment in sepsis. It was shown that children with MyD 88 SNP -938 C/C genotype had more tendency to sepsis in our study. However further studies should be performed for the exact causes of sepsis tendency in a large population.Key Words: Children, MyD 88 gene polymorphism, Sepsis, TLR-9 gene polymorphism.
