Abstract (EN)

In this study, it was carried out to determine the effect of 24-epibrassinolide (24-eBL) and methyl jasmonate (MeJA) on the root growth and secondary metabolite accumulation of adventitious roots in black henbane (Hyoscyamus niger). For this purpose, different concentrations of 24-eBL (0.5, 1 and 2 mg/l) were applied to adventitious roots obtained from leaf stems alone or the combination of 1 mM MeJA for 14 days. After harvest, adventitious roots were evaluated for fresh root weight, dry root weight, root growth index, contents of tropane alkaloids including hyoscyamine and scopolamine and phenolics compounds. As a result of this study, 2 mg/l 24 –eBL was the most suitable application providing the highest root growth parameters while 1 mM MeJA + 2 mg/l 24-eBL combination for scopolamine and catechin and 1 mM MeJA + 1 mg/l 24-eBL combination for hyoscyamine and epicatechin were the optimum applications. On the other hand 1 mM MeJA gave the maximum values for total phenolic, gallic acid, vanillin, cinnamic acid, rosmaniric acid, caffeic acid, chlorogenic acid. Consequently, it was determined that 24-eBL and MeJA treatments can be used successfully in order to increase the production of tropane alkaloids and phenolics compounds in adventitious roots of black henbane.
