Abstract (EN)

Sternbergia fischeriana (Herbert) Rupr. has attractive golden yellow flowers used in ornamental cut flower industry that also contain many medicinally important secondary metabolites. Their large scale cultivation is inhibited as their bulbs grow very slowly and take about 5 years to mature, flower and set seeds. The study aimed to reduce this period through culture of 12 different types of explants i.e. 0.5, 1 and 1.5 cm long two, three, four and five scale bulb explants attached to a thin segment of basal plate. The explants were cultured for bulblet induction on MS medium containing 1, 2, 3, 4, 5 mg/L 2, 4-D or 0.5, 2.5, 4.5, 6.5 and 8.5 mg/L BAP with or without 0.2 mg/L NAA supplemented with 30 g/l sucrose and solidified with 6.2 g/l agar at 15 and 24oC. The results showed that the 2, 4-D failed to induce bulblet regeneration at both temperatures on any type of explant. Variable results were obtained on twelve type of explants containing 2, 3, 4, and 5 scales attached by basal segment cultured on MS medium containing 0.5, 2.5, 4.5, 6.5 and 8.5 mg/L BAP at 15 and 24oC. Maximum number of 4.97 bulblets per 0.5 cm long two scale bulb explant was obtained on MS medium containing 8.5 mg/L BAP-0.2 mg/L NAA. The rooting was obtained on ½ strength MS medium containing 0.19 mg/L NAA. The rooting was affected by the size of bulblet. The maximum bulblet regeneration was recorded on bulblets having diameter of 0.47 cm inducing mean number of 4.33 roots per explant with mean root length of 3.73 cm. The results showed that rooting of bulbs did not take long time depend on bulb diameter.
