Abstract (EN)
Despite recent advances in perinatal management, respiratory distress syndrome (RDS) and its complications are still major causes of mortality and morbidity in newborns. The risk of RDS is inversely related to gestational age. Also genetic, enviromental and acquired factors have important roles in development of RDS. Members of transient receptor potential melastatin are non-selective cation channels expressed in various tissues where they perform diverse functions, such as detection of cold, taste, control of osmolarity, pH, Mg+2 homeostasis, cell proliferation and death. The aim of this study was to determine the association of genetic polymorphisms of TRPM channels with RDS. This study was carried out between the years 2012 and 2013 among the neonates born before the 37th week of gestation and hospitalized in the neonatal intensive care units of city of Gaziantep. A population of 193 premature infants who deveoped RDS and 187 premature infants without RDS was involved in the study. The genetic polymorphisms of TRPM channels were analyzed by polymerase chain reaction (PCR) from the peripheral blood of patients. As a result, T/C ve C/C allele frequencies for rs11070811 and rs74590787 haplotypes of TRPM1 gene; G/C allele for rs9978351 and rs1618355 haplotypes of TRPM2 gene; T/C allele for rs1328142 and rs41452349 haplotypes of TRPM3 gene; AA in rs71352737 (Trp525Ter) polymorphism of TRPM4 gene; C/C/C and C/T/T haplotypes of Arg578Gln, Val254Ala, and Asn235Ser polymorphisms of TRPM5 gene; C/C allele for rs62021060 and rs77165588haplotypes of TRPM7 gene; G/C/C/C/T/C/G, G/C/C/G/T/C/G, G/C/C/G/T/T/G, G/G/C/C/C/C/G, G/G/C/G/C/C/G, G/G/C/G/T/C/G, G/G/T/C/C/C/A, G/G/T/C/C/C/G and G/G/T/C/T/C/A alleles for rs6431648, rs10803666, rs10490018, rs2052029, rs2215173, rs663924 and rs2362294 haplotypes of TRPM8 gene were found to be more frequent in RDS patients when compared to control group. These findings suggest that there might be an association with some genetic polymorphisms of TRPM channels and the increased risk of developing RDS.
