Diagnosis of insulation and molecular diagnosis of Streptomyces which obtained from streams flowing into Van Lake

In this study, isolation, identification and molecular diagnosis of Streptomyces species were aimed from the sediment samples taken from streams pouring into Van Lake. A total of 24 sediment samples were collected from the streams poured into Van Lake. Streptomyces bacteria were isolated from the samples and pure cultures of bacteria were obtained. This resulted in the isolation of 179 Streptomyces bacteria. A total of 77 tests were applied to all the bacteria, mainly color grouping, degradation, nitrogen sources, carbon sources, antibiotics and antimicrobial tests. The obtained results were transferred to IDENTAX Bacterial Identifier 1.2 program and the similarity ratios of the bacteria were determined and diagnosed with the help of computer. According to the results of the major tests, 18.43 % of the species and 35.75 % of the species according to the results of the minors were 95 % and more similar. Test results were entered into MVSP 3.2 computer program and similarity dendograms were created. According to these results, seven minors, four major groups and 16 single members were formed. In addition, sports chain morphology was detected by scanning electron microscopy, which was determined according to color group. 11 Sequence analysis was performed after PCR and electrophoresis of the 16S rDNA gene regions of Streptomyces bacterium. Obtained sequences were compared with species with similar sequences with the help of NCBI-GENBANK. Compared to the 16S rDNA regions, these species were introduced into the MEGA 7 program and a maximum Likelihood phylogenetic tree was established and the phylogenetic positions of the species were determined. According to the results obtained, it was seen that the similarities between 97.82 % and 100 % were obtained when the isolates and the species that they represent are compared.
