Synthesis of complexes, derivative of new thiosemicarbazide ligands and investigation of antitumor properties

Abstract (EN)

In this study; firstly, the thiosemicarbazide derivative ligands were obtained by the reaction different hydrazide and thiocyanate derivatives. These ligands are used in this study; N-(1-Naphtyl)-2-isonicotinoyl hydrazincarbothioamide (L1), 2-(2Hydroxybenzoyl)-N-(1-adamantyl) hydrazine carbothioamide (L2), Isonicotinoyl-N?3-{(4 morpholino)propyl} hydrazine carbothioamide (L3), N-(t-octyl isothiocyanate)-2-isonicotinoylhydrazine carbothioamide (L4). The complexes were synthesized by the reaction of obtained ligands (L1, L2, L3 and L4) with NiCl2.6H2O, CuCl2.2H2O, ZnCl2.2H2O ve CdCl2.H2O, Ni(CH3COO)2.6H2O, Cu(CH3COO)2.3H2O, Zn(CH3COO)2.3H2O, Cd(CH3COO)2.H2O metal salts.The structures of synthesized ligands and their complexes were characterized by using IR, 1H-NMR spectra,13C-NMR spectra, elemental analysis, magnetic susceptibility, UV-Vis., mass spectrum (MALDI-TOF), thermogravimetry analysis-differential thermal analysis (TGA-DTA), and differantial scanning calorimetry techniques.The suggested structures for the L1 complexes of Cu(II), Ni(II) and Cd(II) are octahedral, while Zn(II) complex is tetrahedral. The structures all of the L2 complexes are octahedral, whereas Cu(II) complex of the L3 is square-planar, Ni(II), Zn(II) complexes are octahedral and Cd(II) complex of the L3 is tetrahedral. The Cu(II) complex of the L3 is tetrahedral, octahedral structures are suggested for the Zn(II) and Cd(II) complexes.The cytotoxic effects of the obtained ligands and their complexes were determined by using, one of the enzymatic test methods MTT [3-(4,5-dimetiltiazol-2-yl)-diphenyl tetrazolium bromide] assay method, that is commonly used to evaluate the cytotoxicity. In this study, PC3 prostate cancer cell cultures were used. According to the obtained results, all of the synthesized ligands dose and time dependently reduced cell viability. Between the complexes, all of the L1, L2 and L3 ligands? Cu(II) complexes have led to significant reduction in the cell viability.
