Abstract (EN)
Aim: It was aimed to determinethe frequency and types of both chromosomal anomalies and other genetic elements that cause unsuccessful IVF in patients admitted with a prediagnosis of unsuccessful IVF to examine the relationship between the detected genetic anomalies and the clinical data of unsuccessful IVF patients. Material and Method: Retrospective cytogenetic analyzes of couples who came to Dicle University Medical Biology and Genetics department between 1999- 2019 with a pre-diagnosis of failed IVF will be included in the study. In the pedigree analysis, criteria such as age, gender, consanguineous marriage, smoking and drug use of individuals were considered. Results: In this study, it was observed that 645 cases (76%) out of 850 cases had normal karyotype (46,XX, 46,XY) and 205 cases (24%) had karyotype with anomaly.Although it has seen that 425 couples had at least 1 and at most 6 unsuccessful IVF attempts, considering the factors affecting this situation; Examining the categories such as smokers, drug users, relatives, and those over the age of 35 revealed the existence of some relationships. 317 cases (37%) out of 850 cases smoked, 533 cases (63%) did not smoke. (28%), 306 couples (72%) were found to be relatives. It was observed that 63 cases (7%) of 850 cases used regular medication, 787 cases (93%) did not use drugs, it was observed that 261 cases (30%) of 850 cases were above the age of 35, and 589 cases (70%) were under the age of 35. Conclusion: In conclusion, in our study consisting of patients presenting with unsuccessful IVF prediagnosis, the number of patients with chromosomal anomalies with in the cytogenetic analyzes we made was consistent with the literature, the relationship between the incidence of heterochromatin variants and infertility should be taken into account; therefore, the effects of heterochromatin domains on genes should be investigated in detail and infertility. We think that molecular examination of the approaches directed to this situation will clarify this situation with new studies to be carried out with more patient populations.
