Investigation of plasmid-mediated quinolone resistance genes in acinetobacter isolates

Objective: Acinetobacter isolates are opportunistic nosocomial infections routinely identified in hospitals. These pathogens are mostly isolated from patients in intensive care units. While patients are the primary source of illness transmission from one person to another, medical equipment and hospital personnel are also highly responsible for infection spread. In this thesis study, plasmid-mediated quinolone resistance genes were investigated using the PCR method on Acinetobacter strains obtained from clinical samples, and it is expected that the role of plasmids in quinolone resistance of Acinetobacter species, as well as the presence and prevalence of these genes, will be better understood. Materials and Methods: Between January 2021 and July 2021, clinical samples supplied by the Medical Microbiology Laboratory for standard testing contained 175 Acinetobacter isolates. Following DNA extraction from the Acinetobacter isolates, multiplex PCR was used to test for the presence of quinolone resistance genes. Results: The main goal of our research was to identify the existence of resistance genes that inhibit the activity of quinolone drugs against acinetobacter species. Gene resistance was identified in Acinetobacter species using a total of 175 samples from regular clinical laboratory routines. According to our findings, it was observed that the qnr-S gene was more dominant than other genes. The resistance rates of the genes qnr A, qnrB, qnrC, qnrS, oqxAB, oqxA, and qepA were found to be 0.57, %0.57%, 0.57%, 5.7 %t, 2.8%, 4%, and 0%, respectively.
