The evaluation of the relation of mi-RNA levels between crisis and prognosis of diseases in sickle cell anemia patients who has referred with acute chest syndrome

Abstract (EN)

Introduction and aim: Sickle cell anemia which HbS is in homozigous state is a disorder with frequent crises and results with multiple organ damages. MiRNA are 19-29 nucleotides length RNA's and supress the target gen which is not resulted with translated to protein genome regulation and development, differentiation, and death of the cells. This study, planned to evaluate the relation of miRNA levels in crises and steady state. Method and materials: In this study, blood samples were collected from sickle cell anemia patients in crisis and steady state who were followed up at Çukurova University, Department of Pediatric Hematology. Also, blood samples were drawn from 20 healthy children in the same age group without anemia and systemic disease. İn patients and control groups whole blood count, Hb electrophoresis (acid and alkali pH), and blood chemistry parameters were studied. In patients, hsa-miR 451 and hsa-144 3P were investigated with real time PCR. The parameters were compared statistically. Findings: In sickle cell anemia patients; Hb, hematocrit, and lymphocyte counts were lower than controls, but monocyte and platelet count were higher than controls (p<0.001). In sickle cell anemia patients; Hb A2 % was high, Hb A% was low in the period of acute chest syndrome (p<0.05). Although miRNA-451 and miRNA -144 3P levels were higher than controls in crisis period, it was not significant statistically (p<0.05). Result: The miRNA-451 and miRNA144 3P were high in the period of acute chest syndrome in sickle cell anemia. We need further detailed studies to understand the effect of miRNA on prognosis in sickle cell anemia. Keywords: Acute chest syndrome, Children, microRNA, Sickle cell anemia
