Abstract (EN)
Primary brain tumors are the leading cause of cancer-related deaths with the high incidence rate in childhood. Long noncoding RNAs (IncRNA) are being described as a new class for ncRNA. lncRNAs are associated with various biological roles such as imprinting, epigenetic regulation, apoptosis, cell cycle control, transcriptional and translational regulation, splicing, cell growth, differentiation, and aging. Aberrant expression of lncRNAs are thought to play a critical role in progression of various diseases and development including cancer. In this study; due to the long non-coding RNA expression changes, difference of brain tumor cell lines was aim to determine the role of the molecular pathogenesis. In this context, the expression profiles with human lncRNA and disease related human lncRNA array plate was determined in brain cancer cells and brain cancer stem cell line. U118MG-U87MG; glioblastoma; astrositoma (grade IV), LN18; glioblastoma; glioma (grade IV), Daoy; medullablastoma, SW 1088; astrositoma and human parental brain cancer stem were used. As a control group; human neural stem cells (cortex) and normal human astrocyte cell lines were used. Total RNA was isolated from the cells RNeasy Plus Mini Kit. Reverse transcription procedure was performed for cDNA synthesis by using LncProfiler cDNA synthesis kit. the expressions of disease related human lncRNA and human lncRNA profiles were studied by LightCycler ® 480 real-time PCR. expression profiles of lncRNA genes showed a significant decrease or increase, compared to the control Cells. As a result, fold changes for anti-cancer gene expressions were detected with human lncRNA and disease-related human lncRNA array plates. Data from our experiments indicate that expression profiles of lncRNA may influence tumorigenesis process and that may be useful for additional treatment protocols of human brain tumors.
