Abstract (EN)
Aim: Collagen is a structure in the extracellular matrix and connective tissue that provides mechanical support in the form of triple fibrillar protein. Today it is known that there are 27 different types. Type I, II and III collagen form 80-90% of human collagen. Variables and polymorphisms that can be seen in genes encoding these proteins can cause functional disorders such as excessive elasticity, causing physiological conditions. In this study, we aimed to analyze COL1A1 rs1800012 polymorphism in patients with malocclusion. Materials and methods: In our study, 49 patients with malocclusion were included. The volunteers participated in the study were given detailed information about pre-study analyzes and outputs, and signed informative forms were obtained from them. DNA isolation from buccal swabs by using PureLink DNA isolation kit (Invitrogen, Van Allen Way Carlsbad, CA, USA). Genotyping of all polymorphisms was determined by real-time PCR (RT-PCR) method. Results and conclusion: The number and percentages of CC, AC and AA genotypes for COL1A1 rs1800012 polymorphism were determined as 26 (%53.1),18 (%36.7), 5(%10,2) respectively. When allelic distributions are examined, A and C alleles were found as 28 (28%) and 70 (72%). When the COL1A1 rs1800012 polymorphism was examined in our study group, it was observed that CC genotype and C allele were more dominant. This study shows the possible relationship between upper and lower incisors inclination and skeletal sagittal and vertical relationship phenotypes with the CC,AC and AA genotypes.
