In this study, we aimed to compare the advantages and disadvantages of two different vaginal cytology staining methods in different stages of sexual cycles in female dogs and to find out the reliability of the results of the determined sexual period and fast progesterone test results. In the study material, 37 female dogs were used, and female dogs were divided into three groups (10 dogs in each group) according to their sexual period (prooestrus, oestrus, metoestrus). A group of different female dogs with pyometra was also formed. As a result of smears performed during the progestus period, both erythrocytes and neutrophils were observed in both staining methods. 12.3 ± 2.89% parabasals, 52.4 ± 1.67% intermediates, 27.5 ± 1.23% superficials and 7.8 ± 1.07% keratinized superficial cells with papanicolaou staining method; 10.5 ± 2.7% parabasals, 52.4 ± 1.28% intermediates, 28.4 ± 1.43% superficials with nucleus, 8.7 ± 1.07% keratinized superficial cells were seen by diff quick staining method. Papanicolau staining method when applied during the oestrus period 77.5 ± 0.93% keratinized superficials, 20.3 ± 1.14% superficials with nucleus, 2.2 ± 0.786% intermediary cells; in the diff quick staining method, 78.7 ± 1.08% keratinized superficials, 20 ± 1.06% superficials, 1.3 ± 0.473% intermediary cells were observed and no parabasic cells were observed in both of the methods. Papanicolaou staining method was applied as diuretic stage with 0.8 ± 0.512% keratinized superficials, 11.7 ± 1.12% superficials, 29 ± 1.18% intermediates, 58.5 ± 1.80% basal-parabasal cells; In diff quick dyeing, 1.5 ± 0.885% keratinized superficials, 14% 1.29 core superficials, 28.8 ± 1.19% intermediates, 55.8 ± 1.74% basal-parabasal cells and metoestrus cells were seen in both staining methods. At the same time, a large amount of basal and parabasal type cells, neutrophils and bacteria were seen in smears of group pyometra. The progesterone test kit showed C1-C2 in the proestrous phase, C4 in the estrous stage,C4 in the diestrus stage and C2 scale in the pyometra case. Two different staining methods were compared in terms of cost, ease of use, reliability, image quality of cells. The reliability of the test was determined by comparing the sexual cycle determined by vaginal cytology and rapid progesterone test color scale. The same procedure was applied to dogs with pyrometers. As a result of this study; reliability, advantages and disadvantages were compared by using two different staining methods in vaginal cytology method and it was found that the differentiation of vaginal cells with papanicolaou staining method was easier than diff quick painting method, however, diff quick staining method could be preferred because of its rapid results in clinics.
