The effects of vitamin D on matrix metalloproteinase 9 and apoptosis on experimental rat kidney tissue

Diabetic nephropathy is the most important reason of the end stage kidney disease and it has a major place about the cause of death related to diabetes. Matrix metalloproteinases (MMP) plays a role on disintegration and transformation of extracellular matrix. In this study, it was intended to examine the effects of vitamin D on matrix metalloproteinase 9 and apoptosis on experimental rat kidney tissue. Our study was made with Male Wistar albino rats. They are divided to 5 equal groups which contains 7 members. Any process was made to Control group during 8 weeks. It were given to Buffer group that one dose of phosphate-cytrate buffer intraperitoneally (i.p) and also 50 IU/day vitamin D orally with dropper to Vitamin D group. One dose of 50 mg/kg streptozotocin (STZ) which was dissolved at phosphate-cytrate buffer was given i.p to Diabetes group. Also to the Diabetes+Vitamin D group, after it was given one dose of 50 mg/kg STZ i.p and the next the diabetes develops; 50 IU/day vitamin D was given orally. Rats were sacrified, kidneys were removed at the end of the study. Tissues was burried into paraffin blocks and routine histological follow up was made. İmmunhistochemical tests for MMP 9, TUNEL tests for apopitosis was made to the section of paraffin blocks. Samples were stored at -80 Co for biochemical tests total antioxydant status (TAS) and total oxydant status (TOS). Apoptosis, TAS, TOS, MMP 9 results for the group of Control, Buffer and Vitamin D were the same. When compared with the Control group; TOS, apoptozis and MMP 9 values were seemed increased on Diabetes group althought TAS values were decreased. Comparing with Diabetes group; TOS, apoptosis and MMP 9 values of Diabetes+Vitamin D group were decreased and TAS valuse were increased. In conclusion, our data revealed that it was needed to make further studies to understand the correlation between oxydative stres and MMP 9 on diabetic nephropathy pathogenesis and it can be tested which vitamin D related therapies to prevent the complication of diabetes.
