The effects of epidermal growth factor and octreotide on kidney tissue in experimental diabetes mellitus

In this study we evaluated preventive effects of octreotide and epidermal growth factor (EGF) on the development of diabetic nephropathy in STZ-diabetic rats at the transmission electron microscopy (TEM) and light microscopic level. Male, adult 40 Sprague-Dawley used in this study divided into 5 equal groups as control, diabetic, diabetic+octreotide, diabetic+EGF, diabetic+octreotide+EGF. All rats were sacrificed after 1 month and their kidney tissues were examined in light and electron microscopy. In diabetic group protein deposition in some of the tubular lumens and PAS (+) glycogen deposition with various diameters as vacuole were determined in cytoplasm of tubules epithelium. Increasment in mesangial matrix, narrowing in Bowmann space, obliteration of capillary and pedicell fenestratas, thickening of glomerular basement membran and transformation of microvillus were seen. In octreotide group; increased in mesangial matrix, thickening of glomerular basement membran and decomposed between proximal tubules cells were established. In EGF group; electrodens deposition in mesengium and apoptotic material that were phagocyted by podocytes were seen. In octreotide acetate and EGF group; view were similar to control group In conclusion; combination of octreotide and EGF seem to be more effective in prevention of renal injury in STZ-induced diabetic rats.
