The neuroprotective effect of sildenafil on experimöental spinal cord injury; comparison with methileprednizolone

Spinal cord damage releated to cerebrospinal injuries are still very serious medical issues and cause very serious labour, economical, and social problems. Our goal is to use slidenafilin which is a CGMB inhibitor, proven to treated the erectile malfunction erectile and used as a peripheral vasodilator, to study alternative or supported effect of methyl prednisolone which is used to treat the spinal traumatic patients as a standard treatment.35 Spraque- Dawley rat were used. Group1: (Control group which has Lamiectomy) Group 2: ( Laminectomy + SCI) Group 3: (Laminectomy + SCI + Sildenafile), Group 4: (Laminectomy + SCI + Methyl Prednisolone) Group5 : (Laminectomy + SCI + Sidenafoline + Methyl-Prednisolone) divided as 5 groups which consist of 7 rat at each group. T3-T4 Laminectomy were applied to each and every rat. Aneurysm clips (Yaşargil, 180 gr closing force) were used and hold for 30 seconds to create a damage on spinal cord to each rat at SCI+ teartment groups. 24 hours after application all of the rats in all of the groups were sacrified with high dosage of barbiturate and a section from spinal cord were taken immediatelyfor histopathological evaluation at damaged area by biopsy. In this study, the effects of ischemian that happen after the traumatic spinal cord to histopathological variations after the sildenafileand methylprednisonol teratment were observed. Data we obtained showed that neuroprotective effects of sildenafiline did not show a significant diffences while, according to the Drummond Moore score, motor functions show significant progress. The effects of sildenafiline on spinal cord damages shows a progress on motor functions. However, before using on human there should be more advanced studies releated on side effects and reliable doses on rats.
