Determination of Conservative Effects of Shark Liver Oil and Shark Cartilarge on Oxidative Damage Occurred in Rat Lung Exposed to Formaldehit and Colon Cancer Originated Experimentally. Target: During this study; toxic effects of formaldehite on lung , carcinogen effect of DMH on large intestine , SC and SLO 's conservative effects against these toxic and carcinogic effects has been researched. Methode: 80 rats have been classified as follows : 40 DMH Group: 4 ;control group- 6 ; given DMH but not cured group – 15; given DMH and cured with SC group -15; given DMH and cured with SLO group 40 Formaldehite group : 4; control group – 6;Given FA but not cured group.- 15 ; given FA and cured with SC group -15 ; given FA and cured with SLO group. Tissue samples were taken from the heart of rats in the DMH group for blood biochemical analyzes, tissue samples for pathologic examinations from the large intestine, and tissue samples for storage at -20 in genetic analyzes. Rats in the FA group received blood samples for biochemical analysis from the heart, tissue samples for pathological examinations from the cranial lobe of the right lung, and tissue samples for genotyping -20. In addition, cell culture studies were carried out to determine the therapeutic effect of SC and SLO on colon cancer. CONCLUSION: The adverse effects of DMH could be observed on the rat column in terms of biochemical, genetic and pathological; The adverse effects of FA on the lungs could be evaluated biochemically, genetically and pathologically, and the healing effects of SC and SLO on the colon cancer line could be determined. Kruskal Wallis test was used for the analysis of biochemical values. CA19-9 tumor markers were found to be more elevated in DMH-fed rats than in other markers, and CA19-9 values in DMH + SC and DMH + SLO-fed rats decreased. It can be said that SC is more successful than SLO as a treatment success, since this decrease is found to be more in SC than in SLO. In terms of antioxidant effects, 3 enzymes (CAT, SOD, and GPx) values are increased compared to control in FA application, indicating that administration of both SC and SLO as therapeutics is also reduced in these 3 enzymes compared to control. In comparison with SC, the administration of SLO compared to SC did not cause a significant decrease compared to that of the control group, and the toxic effect of FA application showed that these 3 enzymes showed more antioxidant effect and SC and SLO could be repaired by decreasing the need for defensive mechanism we can say that it has revealed itself. When we look at the MDA values from the biochemical parameters, we see that FA increases in excess amount due to lipid peroxidation in the rats given FA and decreases these values with damage recovery beginning with SC and SLO administration, but these values are slightly above the control group when compared to the control group. When the MDA values were lowered, SC and SLO were about the same level. When genetic results are considered; DMH administration negatively affected the activity of CHOP and EDEM1 genes compared to control. When comparison is made in terms of treatment; The activity of the CHOP and EDEM1 genes was increased, whereas the effect of SC was found to be more successful than the SLO, based on the reduction of ATF4 and ATF6 gene activities. FA application has shown that the CHOP and EDEM1 genes play an active role in the formation of lung damage by reducing the tumor suppressor properties. Based on the reduction of ATF4, ATF6 and GRP78 gene expressions despite treatment with FA for treatment, it is understood that the SLO has more effective in healing effect on lung injury. As a result of pathological examinations of our study, severe lymphoid hyperplasia developed in the group where only DMH was given and colon cancer was tried to be formed. Severe lymphoid hyperplasia is lymphoid hyperplasia in both groups treated with SC and SLO after DMH administration. In cell culture study using Caco-2 cancer cell line and SC and SLO; When the SLO is administered at the 24th hour and 200mg dose; SC means 6 hours and 120mg doses are the most effective way to kill cancer cells. It can be said that our experimental model provides the desired aim and success in this direction. On the other hand, it is believed that colon cancer and lung damage are biochemically, genetically and pathologically illuminated, more cell culture studies are needed in this area, and more extensive follow-up studies are needed to elucidate the treatment options and / or preventable pathways.
