Effects of anti-IL?10 and anti-TGF-ß administration postmating on conception rate in mice.

In this thesis, we were aimed to determine the effects on pregnancy rate and blood serum and uterine fluid concentrations of IL?2, IL?4, IL?6, IL?10, IL?17, IFN?, TNF? and TGFß which have significant effects on pregnancy in females of neutralizing antibodies developed against for IL-10 and TGFß after mating in mice. BALB/C breed study of 21 female mice used in the mating of animals was divided randomly into three groups. The first group was chosen as control group animals. The second group of animals, 0.5 mg of anti-IL-10; a third group animal in the 0.5 mg group of anti-TGFß after mating was injected intraperitoneally. All groups of animals were decapitated and their blood was collected 13th days after mating. Removed from uteri of animals and determine whether they were pregnant. In addition, the animals uterine flushing (irrigation) fluids were obtained. Uterine fluids and blood serum cytokine concentrations were determined using a multiplex immunoassay technique. As a result, pregnancy rates did not differ between the groups. In comparison of cytokine concentrations in uterine fluid of pregnant animals, anti-IL-10 antibody injected TGFß concentration in second group (P<0.01) were found to be higher than other groups. However, other cytokines in pregnant animals found no difference in uterine fluid and blood serum concentrations. Accordingly, it was concluded that the present study in mice after mating anti-IL-10 and anti-TGFß antibodies did not affect pregnancy rates.
