Abstract (EN)

Objective: İn this study our aim is to evaluate the effects of etodolac on kidney, liver and lung with its use in different duration, histologically and biochemically and to observe the effect on tissue of use at different times. Material and Methods: A 3 month old female balb-c mouse was used in this study. Mice were divided into 3 groups; in the first group 8 animals and in the second and third group 10 anilmals. The first group (control) was given 40% dmso by oral gavage. The second group 10 mg/kg etodolac (etol ford 400 mg) was dissolved in %40 dmso and given by oral gavage for 3 weeks. The third group 10 mg/kg etodolac was dissolved in %40 dmso and given by oral gavage for 6 weeks. For biochemical analysis were taken from blood samples from intracardiac. ALT, AST, urea and creatinine levels were measured. Results: Statistically significant differences were observed in group 3 compared to the other groups. İn the liver of group 2 and group 3 occasional sinusoidal dilatations were observed and cell clusters leading to necrosis were observed. İn group 3 necrotic cell covering a large area and cells belonging to the phagocytic system surrounding them were observed. Tubular dilatation and degeneration of tubular epithelium in kidney tissues of group 2 and group 3 were demonstrated with h&E stain. İn addition, a widespread molecular infiltration was observed in group 3. SAA and cd4 antibodies were used in immunohistochemical staining. Amiloid staining with SAA antibody in kidney tissue was shown group 3. Helper T cells in the lung were examined with CD4 antibody. It was observed that itcaused an increase in helper T cells in the lung. Conclusion: In our study, it was observed that the drug had negative effects on the kidney, liver and lung after 3 to 6 weeks of use. More obvious structural changes were observed in the liver and kidney after 6 weeks of etodolac administration. These changes are compatible with biochemical parameters.
