Abstract (EN)
Angiogenesis is a new vessel formation process that plays a role in various physiological and pathological conditions. This process is controlled by the balance between pro-angiogenic and anti-angiogenic mediators in the organism. Angiogenesis is needed for the growth and metastasis of solid tumor. Therefore, the anti-angiogenic treatment approach is seen as an interesting option in cancers. Fenretinide is a synthetic retinoic acid analog and its effects against angiogenesis has been reported in several studies but there are deficiencies in the elucidation of its molecular mechanism. In this study, we aimed to investigate the effects of the fenretinide molecule on some miRNAs and CDH5, FOXM1 and eNOS genes associated with angiogenesis. In addition, the effects of this molecule on HUVEC cell proliferation, cell migration and capillary-like tube formation were shown. In our study, it was observed that the fenretinide molecule applied to the cells in 24 and 48 hour periods suppressed cell proliferation, cell migration and tube formation depending on dose. Total RNA was isolated from cells treated with 1 µM and 5 µM concentrations to investigate the effects of fenretinide on miRNAs and indikatalog noed genes. As a result of simultaneous PCR analysis which is made with these RNAs, it was found that miR-10b decreased significantly and miR-126 increased. In addition, in our study, a significant decrease was observed in the levels of CDH5, FOXM1 and eNOS genes in endothelial cells treated with fenretinide. This study may support the literature to illuminate its molecular mechanism of suppressor roles of the fenretinide molecule on angiogenesis.
