Investigationof TheBiologicalActivity of MethanolicExtract of HazelnutHusk Many diseases such as cardiovascular diseases, diabetes, cataracts and nervous system diseases occur as a result of failure removing free radicals or lack of antioxidants in the organism. The most important source of natural antioxidants are plants and taken into our body with diet. The hazelnu tslag waste (leafy cover) that is surrounding the hard shell of it and by-product formed after blendingis used as an animal bad or burned, so an important part of this waste is not evaluated accordingly. The aim of this thesis is to determine the total phenolic content and antioxidant properties of hazelnut leafy cover. The leafy cover of hazelnut used in the present study was obtained from Bulancak district of Giresun province. The methanolic extract of hazelnut leafy cover was prepared and, the total contents of polyphenolic and flavonoid were estimated by Singleaton, and Salatino and Wosiky, respectively. Activities of FRAB, CUPRAC, reduction potential and DPPH radical cleaning were spectrophotometrically determinated by Benzei and Strain, Apak, Oyaizu and Cuendet methots, respectively. Oxidative damage, induced by exposure of erythrocytes to H2O2 was generated, and the capacitiy to prevent lipid peroxidation with the addition of the extract was studied sepectrophotometrically according to Stockand Dormandy method. The contens of TPFMM and TFMM of the methanolic extract were 128 mg GAE/ g extract and 5.8 mg Quercetin/g extract. Activities of FRAP, CUPRAC, reduction potential and DPPH reduction (IC50) were 173 mg Trolox/g extract, 285 mg Trolox/g extract, 57 mg ascorbic acid/g extract and 0.060 mg/mL, respectively. MDA levels were found to be lower in the group with hazelnut leafy cover (344±79 mg/gHb) compared to the oxidative stres group (824±79 mg/g Hb). According to there sults in the study, it can be considered that hazelnut leafy cover has a high antioxidant feature and preserves this effect in the cellula rmicroenvironment although the it is a waste product. Hazelnut leafy cover isbelieved to have the potential to be industrialistically evaluated as a food additive for different purposes such as medical and cosmetic.
