In this study for the determination of glucose, glucose oxidase enzymes and peroxidase enzymes fixed on the gold electrode by using BSA/gelatin and crosslinking by glutaraldehyde. Measurements were carried out at 0.9 V with amperometric method. During the optimization studies the amount of gelatin, bovine serum albumin and glutaraldehyde were determined. For the optimization working condition pH, temperature, scanning rate and buffer concentration were carried out. Afterwards, some paramaters such as linearity, repeatability of the measurement, shelf life, and serum glucose analyses were carried out. From the bioactive layer optimization studies; gelatin, bovine serum albumin amount and optimal percentage glutaraldehyde were determined as 0.45 gr, 0.030 gr and %2.5 for the Au/BSA-Gelatin/GOD-POD/ glutaraldehyde modified biosensor. Phosphate buffer of pH 7.5 and 50 mM, scanning rate of 0.05 V/s, temperature 35 °C were determined as a optimum working conditions. It was determined that linear results are obtained between the ranges of 5-400 mg/dL for glucose concentration. As for shelf life studies, at the end of 40th day of responses has been preserved by 72%, repeatability of the measurement (n=13) standard deviation (S. S) = ±0,1320 and % coefficient of variation (V. K) = 0.165, respectively. Studies with using serum samples; biosensor, kit, central laboratory results accepted as gold standard ROC curve analysis reviewed and the biosensor results (.90-1.00) were found with excellent range.
