Abstract (EN)

In this thesis study; It was aimed to investigate the presence of IPNV using the Real Time Reverse transcription polymerase chain reaction (rRT-PCR) method in samples taken from 3 rainbow trout farms located on the Eşen stream in Seydikemer District of Muğla Province. For this purpose, 120 maribund rainbow trout weighing 0.5-120 g (30 for rRT-PCR, 30 for histopathology, 30 for bacteriological examination, 30 for parasitological examination) were selected from the enterprises between March and April 2023. For rRT-PCR in infected fish, fish weighing 10 g or less were taken whole into Phosphate-buffered saline (PBS) with sterile antibiotics (10000 IU/ml penicillin + 10 mg/ml streptomycin), and fish weighing more than 20 g were examined for pancreas, kidney, and spleen. and their livers were removed. For histopathological examination, the abdomen of small fish was cut and the liver, pancreas, intestine and gills of large fish were placed in 10% formalin solution. Clinical and autopsy findings in maribund fish were ascites, dark coloration of the skin, exophthalmia, skin lesions, hemorrhage in the eye, anorexia, anemia in the gills, petechial bleeding in the internal organs, kidney wasting, yellow exudate, hemorrhage in the liver, enlarged spleen. As a result of the rRT-PCR study, the presence of IPNV was detected in all 3 farms and in all individuals weighing 0.5-120 g. In the histopathological study, pathological changes were observed in the pancreas, liver, stomach, intestines and gills, especially intense necrosis in the acinar cells of the pancreas and inclusion bodies in the liver.
