Abstract (EN)

Introduction:We aimed to investigate the impact of the curcumin, a natural product, by the histopathological and biochemical examinations in reducing of the ischemia / reperfusion injury as a result of liver blood flow cutting on the liver and distant organs.Recently, Hepatic ischemia-reperfusion models were focused the preventation from the reperfusion injury and its mechanism after liver transplantation and mandatory pringle maneuver during the liver and bile duct surgery. In addition, Hepatic ischemia reperfusion injury doesn?t affect only the relevant organ, affecting other organs especially lungs and kidneys and causes a systematic response in the organism. Therefore, the another goal of this study is to put forward the effects of the same agent on the other organs.Curcumin (diferuloylmethane), the main pigment of Curcuma Longa (turmeric) plant and called a phenolic compound, has anti-inflammatory and antioxidant properties. Curcumin has antioxidant activity, can be compared with Vitamin C and E, increases the endogenous antioxidant defense enzymes, reduces peroksinitrit (OONO-) the formation, expedites the excretion of reactive oxygen radicals and inhibits lipid peroxidation.Material and Methods:We used 40 animals, 10 rat for each group, in our study. Ischemia was applied 30-45 min with the hepatoduedonal ligament clamping and then reperfusion is started. Groups are like these; the first group; only laparatomi, the second group; laparatomi and curcumin, the third group; hepatic ischemia-reperfusion and the fourth group; hepatic ischemia-reperfusion and curcumin.During experimental studies, curcumin capsules were opened and appropriate dose required for animals had been created with the weighing scales. And then the powder was diluted with saline. Fifteen minutes before the ischemia , we gave curcumin through the oral gavage. Sixty minutes after starting the experiment in 1-2 groups, 30 minutes after beginning reperfusion in 3-4 groups (60 minutes after beginning the experiment in all groups), blood was taken from the animals for biochemical analysis and sacrificed in this way. Simultaneously liver, lung and kidney tissue were removed for biochemical and histopathological examination.Results:As a result of the Plasma evaluation; malondialdehyde (MDA) values were higher and antioxidant activity (AOA) values were lower in ischemia/reperfusion group compared with ischemia / reperfusion + curcumin group. The difference between MDA levels were significant (p <0.001), while the difference between the values of AOA were not significant (p> 0.05). As a result of biochemical and histopathological evaluation of tissue samples; AOA values were higher, total oxidant activity (TOA) values were lower and histopathologic scores were better in ischemia / reperfusion + curcumin group, compared with values of ischemia / reperfusion group. However, these differences were not statistically significant (p> 0.05)Conclusion:Curcumin was partly to reduce the effects on hepatic ischemia reperfusion injury on the liver and distant organs, , although this difference was not statistically significant.Keywords: Hepatic ischemia-reperfusion, curcumin, liver, remote organ
