Abstract (EN)

Isolation, expansion, cryopreservation of mesenchymal stem cells obtained from human fat tissue and investigation of differentiation capacities of isolated stem cells. Although there is different isolation, expansion, cryopreservation and differentiation methods used in studies in our country and in other countries, there is no study on animal derived serum free autologous serum and hypoxic culture conditions. Mesenchymal stem cells were obtained from 10 abdominal fat samples taken from 10, 25 to 52-years-old females within the framework of previously defined protocols in surgical conditions. Cell viability, mesenchymal stem cell characterization, freezing / thawing and chondrogenic differentiation in the hypoxic and normal cultured of 1-3-5 passages of the same patient were observed in this study. With this study, it is thought that in the hypoxic culture medium, expansion, renewal and improvement in the subject will be made by adding autologous serum application to existing information about mesenchymal stem cells derived from adipose tissue, and at the same time a standard protocol will be developed and supported by in vivo studies in the future. Working within this context is very important in terms of regenerative medicine application and methodology.
