Investigation of the antitumors effectiveness of tarantula cubensis extract in human hepatocelllular cancer cell culture

Özet (EN)

Background: It is aimed to find out the antitumoral activity of Tarantula Cubensis Extract by suppressing Ki-67 expression on Human Hepatocelluar Cancer Cell Line (HepG2). Material and Methods: HepG2 cells were used in this study. Two groups were formed as HepG2 cell line in the control group and HepG2 cell line to which Tarantula Cubansis Extract (TCE) was administered. First of all, effective TCE cytotoxic and IC50 dose on HepG2 cell line were determined by MTT method. Then, the suppressive efficiency of Ki-67 expression was measured by immunostaining of TCE in HepG2 cell line at IC50 doses determined by MTT method. Results: With the MTT test in HepG2 cell line, the IC50 doses of TCE were determined as 5µg/ml after waiting 24 hours, 1µg/ml after waiting 48 hours and 72 hours. As a result of immunostaining, Ki-67 was detected 75% in HepG2 cells in the control group at the 24th hour, while the suppression rate of Ki-67 expression of 5µg/ml dose TCE was 48%. In the 48th hour control group, 90% of Ki-67 was detected by immunostaining, while the suppression of Ki-67 expression was 35% in the group treated with 1µg/ml TCE. While Ki-67 was detected 92% in the 72th hour control group, the Ki-67 expression suppression was found to be 42% in the 1µg/ml TCE applied group. Conclusion: It was observed that Tarantula Cubensis Extract has statistically suppressed Ki-67 expression in HEPG2 cell line in 24 hours at a dose of 5µg/ml and in 48-72 hours at a dose of 1µg/ml.
