SUMMARY Subject: Evaluation of DNMT1 and HDAC6 expression levels in pregnant women in Intra Uterine Growth Restriction (IUGR) and to shed light on the role of epigenetic mechanisms in the etiopathogenesis of IUGR. Material and Method: A total of 89 pregnant women were included in the study. The pregnant women were classified into three groups as early-onset IUGR (<32 weeks), (n=25), late-onset IUGR (≥32 weeks), (n=25) and healthy pregnant women (n=39) without any additional disease as the control group. Placental DNMT1 and HDAC6 expression were examined by immunohistochemical staining technique. P<0.05 was considered statistically significant. Results: There was no statistically significant difference between the groups in terms of age, gravida, parity and gestational week of birth (p>0.05, p>0.05, p>0.05, p>0.05, sırasıyla). A statistically significant difference was observed between early-onset IUGR and control group (p<0.01), and between early-onset IUGR and late-onset IUGR (p<0.01) in terms of gestational week of birth. There was no statistically significant difference between late-onset IUGR and control group (p>0.05). A statistically significant difference was observed between early-onset IUGR and control group (p<0.01), and between late-onset IUGR and control group (p<0.01) in terms of cytoplasmic DNMT1. There was no statistically significant difference between early and late-onset IUGR (p>0.05). In terms of nuclear DNMT1, a statistically significant difference was observed between the early-onset IUGR and control group (p<0.01) and between the late-onset IUGR and control group (p<0.01). In terms of cytoplasmic HDAC6, a statistically significant difference was observed between early-onset IUGR and control group (p<0.01), between early-onset and late-onset IUGR (p>0.05); There was no statistically significant difference between late-onset IUGR and control group (p>0.05). There was no statistically significant difference between the three groups in terms of nuclear HDAC6 (p>0.05, p>0.05, p>0.05, respectively). A negative correlation was found between DNMT1 nuclear and gestational week of birth (p<0.05). Conclusion: Although no correlation was found between HDAC6 levels and IUGR in placental material, cytoplasmic and especially nuclear DNMT1 levels were observed to be increased in pregnant women with IUGR. A negative correlation was found between DNMT1 nuclear and gestational week of birth Therefore, we think that the increase in DNMTs can be used in the diagnosis of IUGR and/or to improve the prognosis.
