Abstract (EN)
The main purpose of in vitro micropropagation is to propagate plant cells in nutrient medium by providing suitable conditions and preserving their genetic properties. In this study, in vitro compatibility abilities of different Kale (Brassica oleracea var. acephala) genotypes in explant sources and the most suitable explant source were carried out with IAA (Indole-3-Acetic Acid) and BAP (6-Benzyleamino purin) concentrations. A total of three different genotypes obtained from Arhavi, Yusufeli and Rize were used in the research. Root, stem, cotyledon and shoot tip explants of Kale plant were used as explant sources. In the research, 7 different culture media consisting of control MS medium and BAP (0.5 and 1.0 mg/L), IAA (0.5 and 1.0 mg/L) and BAP+IAA (1.0+0.5 mg/L and 1.0+0.1 mg/L) hormone concentrations added to this medium were used. As a result of the research, a general evaluation was made according to the culture media, for the Rize genotype the highest regeneration was 126.4% from the culture medium containing 1.0 mg/lt IAA hormone, and the lowest regeneration was 32% from the culture medium containing 0.5 mg/lt BAP hormone. In the Yusufeli genotype, the highest regeneration was 92% in the culture medium alone, and the lowest was 28.8% in the culture medium where 1.0 mg/lt BAP + 1.0 mg/lt IAA hormones were in balance. In the Arhavi genotype, the best regeneration was 107% in the culture medium containing 0.5 mg/lt IAA hormone, and the lowest was 57% in the culture medium containing 0.5 mg/lt BAP.
