SUMMARY In this research we investigated production of alkaline protease in SSF (Solid- State Fermentation) by Bacillus sp. To understand which is the best for maximum enzyme production we used banana waste, rice husk, millet as substrate for the microorganism. High yields of enzyme were achieved by employing banana waste as substrate, 72 hour incubation time and 1500 urn particle size. Various parameters such as moisture level, inoculum concentration, pH, temperature, carbon and nitrogen sources influencing enzyme production in SSF were optimized. With inhibition study, to characterize the protease, we found that enzyme was inhibited completely by PMSF, which is best known as a serin protease inhibitor. The effect of Ca+2 on enzyme thermal stability was examined. Km value was found to be 3,8. 10"2 for alkaline serin protease.
