Abstract (EN)

Schizophrenia is one of the multisystemic diseases that causes many behavioraland thinking problems. It is a bad ending disease that begins in the childhood and goestill the end of life. Although there are lots of advances in biochemical, anatomical andgenetical areas, schizophrenia is a disease that keeps on staying with having its own lifeand behavioral symptoms and just about by seeing these symptoms it is prognosed.The aim of our study is to determine the numerical or structural abnormalitieswith cytogenetic analysis of 50 patients (25 women,25 men) who were diagnosed asSchizophrenia by using the DSM-IV criteria and show the microdeletion of 22q11region on 22 chromosome by using PCR (Polymerase Chain Reaction) method and todetermine the corelation between them.In cytogenetic analyse of our study lymphocyte culture ang Giemsa-Tripsin(GTG) banding have been done and examined under light microscope.In molecularanalyse DNA is isolated from peripheral blood of the each patient. Target region whichis 185 bp lenght and belogs to COMT gene is duplicated by PCR method usingoligonucleotid primers. PCR product has been performed in %2 agarose gel and imagedby Gel Screening System.The study of determining COMT gene allels by RFLP method (RestrictionFragment Lenght Polymorphism) is targeted as a next part of the study.
