Analysis of survivin gene expressions in extensions in exosomes from breast cancer patients

Abstract (EN)

Breast cancer is an important public health problem for women in the world. Diagnosing patients at an early stage, determining which patients will benefit from neoadjuvant or adjuvant treatment, and determining the risk of recurrence on a patient basis are important factors to improve the survival of these patients. Breast cancer is the leading cause of death in women across the globe. The aim of this study is to determine the role of exosomes and exosomal survivin in breast carcinogenesis. Patients who were diagnosed with histopathologically confirmed breast cancer between 2014-2017 at Medicalpark Gaziantep Hospital Medical Oncology Clinic and healthy volunteers were included in the study. Serum samples from patients and healthy volunteers were stored at -80. Exosomes were isolated from these serum samples. The integrity of the exosomes was disrupted. survivin levels were measured by Enzyme Linked Immuno Sorbent Assay (ELISA). A total of 75 participants, 55 of whom were breast cancer and 20 healthy volunteers, were included in the study. The difference between the two groups was investigated by Mann Whitney U test. There was a significant difference between exosomal survivin levels in the control group (p = 0.047). The exosomal survivin level in the patient group was 2.48 ± 6.38 ng / mL (Range 0-40.452) and in the control group 0.23 ± 0.52 ng / mL (Range 0-2.4). No relation was found between exosomal survivin and other prognostic clinicopathological indicators. It was shown that survivin, which was previously shown as a poor prognostic marker in breast cancer patients, was expressed by exosomal pathway. Exosomal survivin expression is higher in breast cancer patients than in control group. This pathway can be both a therapeutic target and a new marker for early diagnosis of the disease.
