Abstract (EN)

Coupling of the peptides to carrier protein is required to producing of anti-peptide antikor which areweak immunogenic peptides. In aqueuos solution carboxyl grups activation and binding the aminogroups to activated carboxyl group with covalent bond is formed our thesis main subject?sfundamental which is conjugation of globular proteins with synthetic peptides.Carbodiimides are heterobifonctional (zero-length) cross linkers reagents and used for coupling ofcarboxyl groups and amino groups. In this thesis study we prepare Peptide-Protein conjugates whitcarbodiimide method by using 1-ethyl -3-(3-dimethylaminopropyl)carbodiimide hydrochloride. Forconjugation we study with different ratio of peptides for one protein molecule (npeptid/nBSA) andsynthesed the conjugates for this ratio. Conjugates are purified by using different column .Purified conjugates are characterized and the mechanism of the binding process is investigatedcomparatively with synthetic peptide, BSA and BSA-synthetic peptide physical mixture by HPLC,GPC-VISCOTEK (Gel Permeation Chromatography, The system has UV, Refractive Index,Viscometer and Right Angle Light Scattering detectors), Fluoresans Spectrophotometer, Zetasizer (Z-Ave, Zeta Potential). Formation mechanism, electrical charge and three dimensional structure ofbioconjugates are investigated based on physicochemical analysis results.Key words: Peptide, Protein, BSA, Covalent Coupling, Carbodiimide, EDC
