Osteoporosis is a multifactorial systemic bone disease that is described as reduction of bone mineral density, destruction of bone microarchitecture and tendency of fracture with increased fragility. Estrogen deficiency and aging are the main factors for woman osteoporosis. Osteoclast mediated bone resorption dominates osteoblast mediated bone mineralization. The most important trigger of this process is estrogen deficiency because estrogen is one of the most important supressor of bone resorptive cytokins. When we look at the molecular basis of osteoporosis, there have been multiple genes that were investigated in association with bone mineral density. Recently, one of the studied genes of osteoporosis is SOST gene which is localized on 17q12-21 region. We performed a randomized, prospective, controlled study to investigate the possible effect of SOST gene single nucleotide polymorphisms on bone mineral density. The study was consisted of postmenopausal 126 women, with complaint of osteoporosis that was detected with bone mineal density measurement (Group I) and 78 women with high bone mineral density (Group II) who applied to Firat University Hospital, Department of Nuclear Medicine. Rs177885799, rs17886183, rs851054, rs865429 polymorphisms of SOST gene were studied with polymerase chain reaction technique. We determined increased G allele frequency for rs17885799 polymorphism in both groups. For rs851054 polymorphism, we detected 100% variant A allele in both groups. For rs865429 and rs17886183 polymorphisms, we established variant allele exchange, but there was not any significant difference between groups for genotype and allele frequencies.Expanded and different population studies are needed to confirm our results. In our opinion, investigation of SOST gene polymorphisms, which effect bone mineral density, could facilitate to improve new treatment modalities for osteoporosis.Key Words: Postmenopausal osteoporosis, polymorphism, SOST gene.
