Effects of centella asiatica on cell death in prostate cancer cells

Abstract (EN)
Objective: This research was carried out to examine the effect of Centella asiatica on cell death in prostate cancer cells. Material and Methods: The research was carried out to measure the cell death efficiency of Centella asiatica on the DU145 cell line. In our study, the cell death efficiency of the docetaxel agent used in the treatment of prostate cancer and the effects that may occur in combination with Centella asiatica were also analyzed. Cell viability and dose were determined by MTS/PMS analysis for docetaxel and Centella asiatica. Then, the Annexin-V method was used to analyze the membrane change occurring in the early stage of apoptosis, one of the cell death pathways. Control, docetaxel, Centella asiatica and combination groups were treated with the prostate cancer cell line (DU145) and analyzed in triplicate with the Muse Cell Analyzer. Vitality and mortality rates were graphed as percentages by taking their arithmetic averages with the SPSS program. Results: In our study, the effect of Centella asiatica on cell viability did not differ significantly compared to other groups. In early apoptosis, Centella asiatica group increased approximately three times compared to the control group, and the combination group increased approximately two times. In late apoptosis, the Centella asiatica group showed a significant increase compared to the control group, while the combination group increased approximately twice. In total apoptosis, Centella asiatica and combination groups showed an increase more than two times compared to the control group.. Conclusion: In this study, Centella asiatica was found to be a potent apoptosis stimulant in prostate cancer cells and would strongly increase cell death when combined with other chemotherapeutics.
