Virulence genes variation in multidrug resistant pseudomonas aeruginosa isolated from pulmonary infection

Abstract (EN)
Since Pseudomonas aeruginosa is very resistant to antibiotics, more than one antibiotic is usually used in its treatment. This species, which is very common in nature, is abundant in humid environments. They can cause various pathogens such as infection of damaged skin (burns), urinary tract infections (catheters), intravenous infections (injections), meningitis (lumbar puncture), respiratory tract infections (cystic fibrosis pathogens), ecthyma gangrenosum (skin necrosis), otitis externa in swimmers. In this study, samples were collected from 150 patients (40% males and 60% females) with respiratory tract infections. These samples were cultured on different culture media such as MacConkey agar and Cetrimide agar and identified by biochemical tests. After identification, P. aeruginosa isolates were stored in LB broth supplemented with 20% glycerol at -20°C for later use. The results obtained showed that all samples isolated from patients were positive for bacterial growth on optimal culture media such as agar medium and mannitol saline agar. Moreover, positive bacterial growth was detected in 35 (23.3%) of the samples. Furthermore, PCR products were resolved by gel electrophoresis and specific amplification of bacterial CTXM (676bp), GES (692bp), OXA (928bp), TEM (766bp) and SHV (827bp) genes of the expected size was confirmed. In addition, Pseudomonas aeruginosa showed resistance to different antibiotics. While the highest resistance was determined against Ceftazidime, the lowest resistance was determined against Ciprofloxacin. It was concluded that Pseudomonas aeruginosa bacteria obtained from different clinical sources were resistant to many antibiotics. The highest resistance was found against Ceftazidime and the lowest resistance was found against Ciprofloxacin. The results revealed that Pseudomonas aeruginosa was resistant to many antibiotics.
