Radiolabeling formulation of temozolomide loaded solid lipid nanoparticles and investigation of radiopharmaceutical potential

Abstract (EN)
Recently, 99m Tc labeled compounds are extensively used for imaging of target organ. In this study, an anticancer agent temozolomi de loaded solid lipid nanoparticles synthesized and they radiolabeled with [ 99m Tc(CO ) 3 ] + core. TMZ - SLNs morphological characteristics, particle size and zeta potential were determined by Dynamic Light Scattering (DLS), Zeta Potential Measurement (ZP) and Scanning Electron Microscopy (SEM) methods. The particle size by DLS analysis is 120 ±20 nm and the value of the zeta potential is - 30,4 mV and the particle size using SEM images is observed in varying sizes between 80 - 200 nm in spherical morphology. The entrapment efficiency of the solid lipid nanoparticles is determined as 49.25%. After the quality control studies, t he radiolabeling efficiency of [ 99m Tc(CO) 3 ] + core radiolabeled temozolomide ( [ 99m Tc(CO) 3 ] - TMZ) was 99. 3 % ±0. 35 and the radiolabeling efficiency of temozolomide loaded solid lipid nanoparticles ( [ 99m Tc(CO) 3 ] - TMZ - SLN ) was 90 . 04%± 0.3 . In this study, activites of solid lipid nanoparticles on cells is investigated on U87 MG ( human glioblastoma - astrocytoma ) and Daoy ( human medulloblastoma ) cell lines. IC 50 values in 48 hours of TMZ is determined on U87 MG and Daoy x cell lines as 1241 μ m and 501 μm respectively. The study of t he changing of the binding activity by time on cells , the greatest binding efficiency was determined in 240 minutes for [ 99m Tc(CO) 3 ] - TMZ - SLN as 33,36 %±3.77. In conclusion, obs erving biological behavior of solid lip id nano particles of in vitro studies is obtained by forming radiolabeled an anticancer drug loaded solid lipid nanoparticles.
