In order to restore the immundisregulationdeveloped by surgical trauma, sveveral methods are being tried. Aim of this study isto investigate the effects of beta-glucan on peritoneal immune system and cytokineresponse in rat pneumoperitoneum model.Twenty eight rats were divided into three groups like; group 1, n=9 (sham- notoperated); group 2, n=9 (control, 10 mmHg pneumoperitoneum for 30 min); group 3,n=10 (beta-glucan, 100 mg/kg/day beta-glucan by gavage, for 7 days beforepneumoperitoneum). 24 h after pneumoperitoneum, samples of peritoneal fluid wascollected from all rats and CD4, CD8, CD25 T cells counted bt flowcytometry andlevels of IL-4,IL-10, IL-12, IFN-gamma, TGF-beta measured by ELISA.CD4+CD25+ T cells (Treg) were significantly higher in beta-glucan group fromothers and IL-10 levels were significantly higher in beta-glucan group whencompared to sham (p<0.05). IFN-gamma was higher in control group whencompared to sham and it significantly decreased in beta-glucan group compared withcontrol (p<0.05). There was no difference in CD4+ T cell ratio between all groups butCD8+ T cells were slightly higher in beta-glucan group. TGF-beta was higher in beta-glucan group than others. There was no significant difference in IL-4 and IL-12among all groups.Pneumoperitoneum triggers a peritoneal immundisregulation but beta-glucantreatment reduces the degree of this disregulation by increasing regulatory T cellsand suppress the developing inflammation. This is the first study in the literaturewhich investigates the effects of beta-glucan in experimental laparoscopic surgeryand regulatory T cells. For this reason, to investigate the effects of beta-glucan onthe other components of immune system and to determine the effects developing bydifferent dosage and usage period of this drug further investigations are needed.
