Stem cells are undifferentiated cells with the potential to self-renew and can change into many specialized cell types in a proper milieu. Because it was discovered that mesenchymal stem cells, one of the stem cell kinds, develop not only into the mesenchymal layer but also into tissue cells that differentiate from layers other than it, mesenchymal stem cells have become the most preferred stem cell type in research. Mesenchymal stem cells can be obtained from many places such as bone marrow, adipose tissue, dental pulp, and placenta. Mesenchymal stem cells originating from adipose tissue can be easily obtained in large quantities and are as effective as hematopoietic stem cells. These characteristics allowed us to select adipose tissue for our research. In our study, the relationship of kisspeptin and neurokinin gene expressions of mesenchymal stem cells obtained from rat adipose tissue with the number of passages was examined. First, mesenchymal stem cells obtained from rat adipose tissue were grown in cell culture medium until passage 10. Mesenchymal stem cells were determined by checking whether the cells expressing CD90, CD45, CD29 surface antibodies in flow cytometry. Finally, after RNA isolation of stem cells and cDNA reverse transcription, gene expression of Kiss-1, Kiss1R, TAC2, TACR2 genes in different passages were examined. . As a result, we observed that the Kiss-1 gene was not expressed in adipose-derived mesenchymal stem cells, but the Kiss1R, TAC2 and TACR2 genes were expressed, and the expressions of these genes changed a lot according to the number of passages.
